ロサンジェルス（ニューヨークの説もある）でソーシャルワーカーと不動産業をしている両親の元で生まれ育ったリサ・ゲイ・ハミルトンは、3歳にして演劇を始める。3年生の時にはコメディアンのフィリップ・ウィルソンを演じていた。中学校に入ると演技のプライベートレッスンを受けていた。ニューヨーク大にてシアターの学位を取得、その後にジュリアード大もシアターの学位を取っている。
映画には1985年の「Krush Groove / クラッシュ・グルーブ」の小さな役でデビュー。1993年のニューヨークのシェイクスピア祭にてケビン・クラインと共演した舞台「Measure for Measure」のイザベラ役で有名になる。その頃から映画への出演も増えた。
